I just use Aseprite. It’s not free but cheap and really good. I export to PNG files and convert them to screen 5 bitmap data with a conversion script I wrote in Node.JS. I wouldn’t use some homebrew editor dedicated to screen 5 (if it exists), I doubt it’ll be as feature-rich as a modern general purpose pixel art editor.

Just make a 256x212 image in indexed mode with a 16-colour palette. The only thing you need to pay attention to is that you pick the right colour component values: 0, 43, 82, 118, 153, 188, 222, 255. I was thinking to make a little Lua script which adds a button to automatically quantises the colours, but haven’t gotten around to that yet.
